Genetic Characteristics of Citrus Tristeza Virus Isolates from Cultivated Citrus in China Based on Coat Protein Gene

چکیده

Citrus tristeza virus (CTV) is an important citrus pathogen causing considerable economic loss to production. Knowledge on genetic evolutionary of the CTV population in China remains limited. In this study, 1439 samples were collected from nine citrus-producing areas China. The coat protein (CP) genes amplified by RT-PCR, and sequenced analyze evolution. Analysis base composition showed AU preference pattern, with GC content was lower than content. Nine populations clustered into one clade neighbor-joining (NJ) tree, indicative a close phylogenetic relationship among molecular variation (AMOVA) revealed that 77.72% variations observed populations, F ST value 0.223. values dN/dS neutrality test CP gene ranged 0.016 0.082 ?1.377 1.456, respectively, results suggesting all relatively constantly maintained under purifying selection. Our study demonstrated characteristics evolution China, provided theoretical basis for scientific control CTV.

Cloning and expression of coat protein gene of the most prevailing citrus tristeza virus isolate in Northern Iran

Citrus tristeza is one of the most destructive diseases of citrus and its causing agent CTV was introduced from Japan to northern Iran in the late 1960s. After about three decades of restriction to originally infected Satsuma trees, natural spread of CTV has being undertaken and melon aphid (Aphis gossypii Glov.) was confirmed as a potent vector in these areas. Regarding the specificity of aphi...

Characterization of the Full Length Coat Protein Gene of Iranian Grapevine fanleaf virus isolates, genetic variation and phylogenetic analysis

The full-length coat protein gene of Grapevine fanleaf virus (GFLV) isolates from Iran was characterized byreverse transcription polymerase chain reaction (RTPCR) and sequencing. The expected 1515 bp coatprotein (CP) gene amplicon was obtained for 16 isolates out of 89 that were identified by double antibodysandwich enzyme-linked immunesorbent assay (DASELISA) in a population ...
